A bunch of really smart people at the GPS software firm Geotab just did the math and put together an interactive map that showcases the least-traveled routes in each U.S. state.
In Colorado it is U.S. Highway 385, also known as the High Plains Highway. Which according to Wikipedia is north of Cheyenne Wells, and is the easternmost significant north–south state highway in the state of Colorado. It enters the state from Oklahoma while overlapped with US 287, but splits at Lamar to follow its own route through the Eastern Plains to Nebraska.
